Hmmmm.....I just tried to change a user's permissions and got the same "Ajax Reply Error".

Please PM backend SA login credentials so may take a look. I never received your credentials so was not able to review your install. Please keep in mind SA access is required so debug settings can be adjusted in global configuration.

Have created a bug ticket to have your issue further infestigated. Unsure what the cause is directly, but appears to be some sort of issue with the ajax checking. Appears only present when editing a user other then your self.
